Is there a difference in power when dynoing in certain gears?

Just curious.  My bike was just tuned and they did all the power dyno in 4th gear, streetability in 3rd gear, and checked for detonation in 5th gear?
The tuner said that's what the people at Dynojet taught him to do.
Sound right?

I forgot to add:  Bike is a 07 StreetGlide (6 spd)
